:甲















ヒトの舌背部には膨大な数の口腔微生物が安定な常在微生物叢を構築して生息している。この舌
微生物叢の優占種は多くの人に共通である一方、各優占種の構成比率は人それぞれ異なる。本研究
では唾液に含まれ口腔細菌の増殖への影響を及ぼすことが知られている酪酸に着目し、舌苔微生物
叢の細菌構成との関連について検討を行った。まず69名の成人男性から採取した含嗽液に含まれる
酪酸濃度を測定するとともに次世代シーケンサーを用いて16S rRNA遺伝子群を解読する網羅的細
菌群集解析法により舌微生物叢の細菌構成を決定した。その結果、含嗽液に含まれる酪酸濃度の高
い者ではPrevotella histicola、Veillonella atypica、Streptococcus parasanguinisの構成比率が高く、
Neisseria subflavaおよびPorphyromonas pasteriの構成比率が低いことが示された。続いて異なる濃度
(0、0.8、1.6、3.2 mM)の酪酸ナトリウムを添加したBasal medium containing mucin (BMM) 培地に
て12名の成人から採取した舌苔微生物叢を培養し、舌常在微生物の増殖に酪酸が及ぼす影響を検証
した。その結果、培養後の細菌構成に添加に伴う統計的に有意な差異を認め、培養後の細菌群集に
おけるNeisseria subflavaの構成比率が減少することが示された。以上の結果から酪酸が部分的にでは
あるが舌苔微生物叢の構成バランスの悪化に寄与していることが示唆された。
