Studies on extrinsic effects on the pattern formation via Delta-Notch signaling pathway
概要
Here, I start from liver histology. The liver is composed of hepatocytes that produces bile
and cholangiocytes that transports bile. The differentiation of cholangiocyte occurs from
cholangiocytes differentiation during mouse embryonic days 13.5 (E13.5) and E18.5
(Gordillo et al. 2015) around the portal vein in the liver (Crawford et al. 2010). During
this period, Delta-Notch signaling pathway is critical in the differentiation to
cholangiocytes from progenitor cells (hepatoblasts) (Hofmann et al. 2010).
This signaling pathway is composed of two constituents: Notch receptors and Delta
ligands. Notch intracellular domain (NICD) leaves from the receptor and act as a
transcription factor, which supresses Delta synthesis (lateral inhibition), when the
interaction of Notch receptors and Delta ligands at the opposing cell surfaces occurs
(trans-interaction), (Axelrod. 2010). In addition, overexpression of NICD causes the
expression of the markers for cholangiocytes in vitro (Tanimizu and Miyajima. 2004). In
addition, recent studies suggested that Notch receptors and Delta ligands on the same cell
membrane bind to inactivate each other (cis-interaction) that shortens the time until the
equilibrium (Sprinzak et al. 2010) (Figure 1). Since these interactions were critical in
many developmental processes (Sprinzak et al. 2010) (Collier et al. 1996) (Miller et al. ...
