Analysis of the activation mechanisms of Sirt1 by KPMF-8 and GAPDH
概要
This study revealed the molecular mechanisms of Sirt1 activation by KPMF-8, a natural compound that is much more potent than resveratrol, and by GAPDH. Both KPMF-8 and GAPDH have effects on the relative orientation of the NTD and CTD of Sirt1. Although this effect by KPMF-8 is dependent on the presence of the substrate peptide, that of GAPDH is independent from the substrate peptide. The results obtained in this study will contribute to the control of the activity of Sirt1, an important regulator of ageing.
