総括および今後の展望
4.1. 総括
本研究では、放射線照射時に誘発される初期の生物学的影響である DNA 損傷、と
くに染色体異常や細胞死といった重篤な損傷を誘発しやすい DSB に着目した。そし
て、放射線照射後の物理学的過程(電離・励起の発生)について MC コードを用いて
解析し、初期の生物学的過程である DNA 損傷生成率を推定した。
第 2 章では、細胞核内に発生する DSB を計測することが可能な γ-H2AX focus 形成
法と、MC コードによる電離、励起の局所密度解析とを組み合わせることにより、光
子線照射時の complex DSB 生成率を簡便な実験手法にて推定することに成功した。γH2AX foci 面積とナノメートルスケールの領域内に発生する電離、励起数に比例関係
が成立すると仮定し、complex DSB の生成に必要な電離、励起数の閾値を γ-H2AX foci
面積の閾値に換算した。その結果、γ-H2AX foci 面積の閾値から各 X 線エネルギーの
complex DSB 生成率を推定することが可能となった。加えて、光子線エネルギーの生
物学的影響を考慮する際は、complex DSB 生成率ではなく、総 DSB 数が重要である
ことが明らかとなった。
第 3 章では、現在普及の進む MRgRT を想定した電子線照射の生物学的影響に対す
る磁場の影響を、MC シミュレーションを用いて DNA 損傷生成率を解析することで
評価した。放射線治療で用いられる高エネルギー電子線(1 MeV, 10 MeV 電子線)で
は、線量分布や電子線飛程のようなマクロスケールの領域で磁場強度の増加により変
化が確認された。一方で、ナノメートルスケールでの電離、励起密度を反映する DSB
生成率では磁場による変化が確認されなかった。したがって、MRgRT を行う際の治
療計画では、磁場による線量分布を考慮する必要はあるが、DSB 生成率すなわち生物
学的影響は、従来の非磁界下における放射線治療と同等とみなすことが可能であるこ

とが示唆された。
これらの研究では、MC シミュレーションを用いることで、complex DSB 生成率の
新たな実験的解析手法の開発および新たな放射線治療における生物学的影響の推定
を遂行した。本論文で扱われた 2 つの研究より、MC シミュレーションの利用は
complex DSB のような細胞実験のみでは解析不可能である損傷の精細な解析が可能と
なる。また、磁界下における電子線照射といった、実測可能な施設が限られるような
体系をシミュレーション上で模擬し、新たな放射線治療を行う場合の DNA 損傷生成
率を推定することが可能となる。これらの研究から、MC シミュレーションを用いた
物理学的過程(電離・励起の空間分布)の解析により、治療に資する生物学的過程の
初期応答(DNA 損傷生成率)の高精度な推定に成功したといえる。
4.2. 今後の展望
本研究にて行われた解析の高精度化を図るために、次のような事項が今後の課題と
して挙げられる。
本研究の DSB 生成率解析に用いられた MC コード(PHITS および WLTrack)は、
物理学的過程(二次電子線の飛跡構造)のみ考慮している。したがって、二次電
子線が誘発する電離、励起により直接引き起こされる DSB のみを定量化してお
り、ラジカルの挙動(拡散、相互反応)とそれによって引き起こされる DSB は推
定しておらず、化学的過程については考慮していない。本研究では電子線・光子
線といった低 LET 放射線を使用しており、電離・励起の空間分布がシングルスパ
ーのような離散的分布であるため、ラジカルの反応がスパー内で完結する。その
ため、直接作用による DSB 生成率と間接作用による DSB 生成率が比例関係とな
り、DSB 生成率の相対値には影響を及ぼさない。しかし、高 LET 放射線のような
飛跡に沿って多数の二次電子線が発生する線質の放射線を解析する場合は、ラジ
カルの反応もさらに考慮する必要があると考えられる。このように、様々な放射
線の種類において生物学的影響をより正確に把握するためには、化学的過程の解
析が可能なコードの開発が必要である。

総括および今後の展望
本研究では、生体内の構造とは異なる任意体系でのシミュレーション解析・細胞
実験を行っている。シミュレーションにおいては、液相水中における電子線飛跡
構造解析にて DSB 生成率を推定したが、実際の生体では、液相水と比較して、よ
り不均一かつ複雑な構成であると考えられる。また、臨床適応となる悪性腫瘍に
関しては、細胞や細胞核等の大きさが不均一である。そういった腫瘍内不均一性
においてもシミュレーション解析を可能とするためには、将来的に生体構造をよ
り正確に模擬できるような体系をコード内に組み込み、検証する必要がある。細
胞実験においては、より実際の生体構造に近い 3D 培養法や in vivo 実験への応用
について検討する必要がある。
【第 2 章 電子線飛跡構造解析に基づく複雑な DNA 損傷生成の実験的解析】
本研究では、実験的解析のみで完結する手法にて complex DSB 生成率を実測して
いない。今後の課題として、SSB や BD に対する免疫蛍光染色を行い γ-H2AX foci
との共焦点を観測するといった手法を用い、complex DSB の実測値を取得し、本
研究にて開発された解析手法による complex DSB 推定値との比較が必要である。
本研究にて開発された complex DSB 生成率推定手法は、光子線や電子線といった
低 LET 放射線のみを対象としている。低 LET 放射線照射時に生成される γ-H2AX
foci は隣接する foci 同士が重なる確率が低く、foci 面積が DSB の複雑さを反映し
ている。しかし、高 LET 放射線では、飛跡に沿って大量の二次電子線が発生する
ため、foci 同士の重なりを考慮する必要がある。今後は高 LET 放射線にも適用可
能となるよう、解析手法の発展が期待される。
【第 3 章 MRI 融合放射線治療にて磁場が飛跡構造および DNA 損傷生成率に与える
影響の推定】
本研究にて解析したエネルギーと同一の電子線照射による実測データは存在せず、
本研究にて解析された DSB 生成率は実測値と比較されていない。etsmode による
推定値は、先行研究にて良く検証された EGS モードの推定値と良い一致を示し、

妥当性は検証されている。しかし、今後の課題としてシミュレーションと同条件
での照射実験による DSB 生成率の測定値を取得し、MC コードによる推定値と比
較検討する必要がある。
